Stipple Bio Raises $100 Million Series A to Advance Precision Oncology Platform and Cancer Drug Pipeline
Stipple Bio, a Cambridge, Massachusetts-based biotechnology company developing precision oncology therapies, has raised $100 million in Series A financing to advance its lead cancer therapy candidate and expand development of its proprietary drug discovery platform. The oversubscribed financing was co-led by RA Capital, a16z Bio+Health, and Nextech Invest, with participation from existing investors including Emerson Collective Investments through Yosemite, GV, LoLa Capital Partners, and GordonMD Global Investments.
Founded in 2022 by cancer biology researchers Dr. Aaron Ring and Dr. Aashish Manglik, Stipple Bio is focused on developing next-generation precision oncology medicines by identifying tumor-specific cell surface epitopes. The company’s technology platform, known as Pointillist, is designed to map and identify highly specific targets on cancer cells that can be used to create therapies intended to selectively attack tumors while minimizing damage to healthy tissues.
The newly raised capital will be used to advance Stipple Bio’s lead therapeutic candidate, STP-100, into early clinical studies and further develop the company’s oncology pipeline. The company said the financing is expected to support operations through 2029 as it continues building medicines based on its epitope-level approach to cancer treatment.
Stipple Bio’s Pointillist Platform is built around the concept that precise targeting at the epitope level can improve the effectiveness and safety profile of cancer therapies. While many oncology approaches focus on identifying tumor-associated genes or proteins, Stipple Bio is focused on finding specific regions on cell-surface molecules that distinguish cancer cells from normal tissues. The company believes this approach could help expand the number of targets available for precision oncology drug development.
The company’s lead program, STP-100, is an antibody-drug conjugate (ADC) designed with tumor-specific binders intended to reduce on-target, off-tumor toxicity. Stipple Bio expects the candidate to enter early clinical studies in 2027 as it works to evaluate its potential as a targeted cancer therapy.
RA Capital participated as a co-lead investor in the Series A financing. The investment firm specializes in life sciences and healthcare companies, supporting biotechnology businesses developing innovative therapies and medical technologies. RA Capital’s participation reflects growing investor interest in precision oncology platforms that aim to improve therapeutic outcomes through more selective targeting approaches.
a16z Bio+Health also co-led the financing. The investment platform from Andreessen Horowitz focuses on biotechnology, healthcare, and life sciences companies developing technologies with the potential to transform medicine. The firm previously supported Stipple Bio’s early development and continued its backing through the Series A round.
Nextech Invest joined RA Capital and a16z Bio+Health as a co-leader of the investment. Nextech Invest focuses on biotechnology companies developing breakthrough treatments, particularly in areas such as oncology and precision medicine.
Additional investors participating in the financing included Emerson Collective Investments, managed through Yosemite, GV, LoLa Capital Partners, and GordonMD Global Investments. These investors have supported Stipple Bio’s development as the company works to advance its precision oncology technology and therapeutic pipeline.
Stipple Bio’s latest financing builds on earlier support from investors who backed the company during its early stages. The company previously received seed financing from investors including Andreessen Horowitz, Emerson Collective, GordonMD Global Investments, GV, LoLa Capital Partners, Magnetic Ventures, and OMX Ventures.
The funding comes as pharmaceutical companies and biotechnology startups continue searching for more precise cancer treatments. Traditional cancer therapies often face challenges related to toxicity because treatments may affect healthy cells alongside cancer cells. Precision oncology companies are attempting to address these limitations by developing therapies that more specifically recognize and target malignant cells.
Stipple Bio’s founders believe that tumor-specific epitopes represent an important opportunity for discovering new cancer targets. The company’s technology combines cancer biology, molecular profiling, and drug development expertise to create therapies designed around highly selective tumor recognition.
The company is also expanding its leadership and scientific capabilities as it moves toward clinical development. Alongside the financing announcement, Stipple Bio announced that Derek DiRocco, PhD, and Thilo Schroeder, PhD, would join its board of directors.
With the $100 million Series A financing secured, Stipple Bio plans to accelerate development of STP-100, expand its Pointillist Platform, and identify additional oncology programs. The company aims to establish a pipeline of precision medicines that use tumor-specific epitope targeting to create more effective and safer cancer therapies.
As biotechnology investors continue prioritizing next-generation oncology approaches, Stipple Bio is positioning itself as a company focused on solving one of cancer treatment’s central challenges: delivering powerful therapies while minimizing harm to healthy tissue. Through its precision targeting platform and expanding investor support, the company is working to advance a new generation of cancer medicines.
